nervous tissue or in the incubation medium in the presence of L-leucine should be considered as a direct evidence of DA synthesis by monoenzymatic TH neurons in cooperation with any other neurons, possessing AADC. The L-DOPA concentration in incubation medium was 0,027 ± 0,003 ng/ml. It was maintained at the same level after incubation in KrebsRinger solution with L-leucine (0,033 ± 0,003 ng/ml). However according to our hypothesis incubation with leucine should not only decrease DA levels but also increase L-DOPA levels in the incubation medium. This inconsistency could be explaned this way: both L-DOPA and DA are metabolized by COMT, therefore when DA levels decrease this could attenuate L-DOPA affinity to COMT, increasing its degradation via this enzyme. According to this assumption COMT inhibition should cause LDOPA increase in incubation medium with L-leucine in comparison with the medium without L-leucine. In order to test this hypothesis, we have performed a second series of experiments by incubating the MBH initially in Krebs-Ringer solution with tolcapone, an inhibitor of COMT, and then in the same solution containing or lacking L-leucine. SN was used as a control. The L-DOPA concentration in incubation medium of the Krebs-Ringer solution, containing tolcapone and L-leucine was significantly higher than in Krebs-Ringer solution containing only tolcapone - 0,14 ± 0,04 ng/ml vs. 0,23 ± 0,03 ng/ml. Thus, the inhibition of the cooperative synthesis with L-leucine perfusion decreases DA levels by 40% both in tissue and in medium and increases L-DOPA levels by 39% in the medium, that conclude the significant role monoenzymatic neurons in the DA synthesis in MBH of adult rats.
