human duodenal mucosa biopsies [1], we have shown for the first time that cathepsin G is synthesized not only by the innate immune cells (neutrophils, mast cells, monocytes), but also by epithelial cells located in the bottom of intestinal crypts Paneth cells (PC). Cathepsin G-specific immunofluorescence was detected in the PC zone of rough endoplasmic reticulum, in the area of secretory granules and in the lumen of the intestinal crypt [1]. PC secretes into the lumen of the intestinal crypt the antimicrobial factors which protect stem cells of the intestinal epithelium against damage by pathogenic microorganisms. Our results about the immunolocalization of cathepsin G indicate constant presence of this protease in intestinal epithelium area, which can make significant adjustments in understanding of cathepsin G role in the intestinal mucosa physiology. It was established that goblet cells are the source of renin biosynthesis in rat intestinal epithelium [4]. It is known that renin, located on top of the RAS activation cascade, is synthesized as an inactive precursor - prorenin, activation of which may occur by proteolytic processing of cathepsin G [3]. Data presented in this paper about cathepsin G localization together with above mentioned published data give reason to assume the existence of cathepsin G-dependent pathway of local RAS activation in small intestine, where cathepsin G plays a key role in initiating the emergence of active AT-II in the intestinal epithelium.
